A Danish cohort study of 1,180,308 singleton children investigated whether paternal valproate use during spermatogenesis increased neurodevelopmental disorder (NDD) risks in offspring. Prompted by a conflicting European Medicines Agency safety report, researchers compared 961 children exposed to paternal valproate monotherapy against 1,401 children exposed to lamotrigine or levetiracetam. The main analysis revealed no significant association with NDDs (adjusted hazard ratio, 1.02). This safety profile remained robust across various secondary and sensitivity adjustments. Ultimately, the findings suggest paternal antiseizure medication exposure during conception poses no major risk to offspring, though researchers emphasised a need for full study transparency.
